The Isolation,Purification and Some Properties of Two Toxins From the Scorpion Venom of Buthus martensii Karshi From Liaoning

  • The crude venom of scorpion Buthus martensii Karshi (from Liaoning) was fractionated into 12 protein peaks on CM-Sephadex C-50,Two toxins were purified from the eighth fracton by rechromatography on CM-Sephadex C-50 and gel filtration on Sehadex G-50.Their homogeneties were demonstrated by polyacrylamide gel electrophoresis,isoelectric focusing electrophoresis and SDS-polyacrylamide gel slab electrophoresis.The molecular weight of the toxins,calculated from measurement in SDS-Discontinuous polyacrylamide gel slab electrophoresis,are 8,980 and 8,660 respectively.Their isoelectric points are 7.58 and 7.90 respectivey by isoelectric focusing electrophoresis.The amion acid composition of the toxin Ⅰ was determined.The crude venom showed hyaluronidase activity but on phospholipase A2,α-amyase,arginine esterase,acetylcholinesterase,phosphomonoesterase,L-amino acid oxidase as well as proteinase activity.The LD50 of crude venom of scorpion Buthus martensii Karshi was found to be 10.3 μg/g for mice.The protein of molecular weight less than 10,000 dalton could be determined by SDS-disc polyacrylamide gel slab electrophoresis (13%).The results obtained were more satisfactory.
