Modeling postpartum depression in rats: theoretic and methodological issues

  • 摘要: 产后期是指在分子、细胞、生理和行为水平发生一系列改变,为女性的母性行为表达做准备的一个阶段。这些正常适应性变化的改变或抑制会导致情绪、动机和认知能力调节的异常,引起产后精神障碍,比如产后抑郁。尽管这种疾病发病率高,对母亲和孩子不利,但是其病因和相关的神经生物学机制仍然知之甚少,部分原因是由于缺乏适当的动物模型。近几十年来,已经有一些在大鼠上的尝试性研究来模拟产后抑郁。在本文中,我们首先描述了产后抑郁症的临床症状,并讨论已知的危险因素,包括遗传和环境方面。此后,我们讨论了多个已经建立的大鼠模型和这些尝试中获得的知识,而这些模型涉猎了产后抑郁的多个方面。在此过程中,我们关注每一次研究背后的理论和实现自己目标的方法。最后,我们提出了几个可研究领域,并为未来的发展方向提出建议。

     

    Abstract: The postpartum period is when a host of changes occur at molecular, cellular, physiological and behavioral levels to prepare female humans for the challenge of maternity. Alteration or prevention of these normal adaptions is thought to contribute to disruptions of emotion regulation, motivation and cognitive abilities that underlie postpartum mental disorders, such as postpartum depression. Despite the high incidence of this disorder, and the detrimental consequences for both mother and child, its etiology and related neurobiological mechanisms remain poorly understood, partially due to the lack of appropriate animal models. In recent decades, there have been a number of attempts to model postpartum depression disorder in rats. In the present review, we first describe clinical symptoms of postpartum depression and discuss known risk factors, including both genetic and environmental factors. Thereafter, we discuss various rat models that have been developed to capture various aspects of this disorder and knowledge gained from such attempts. In doing so, we focus on the theories behind each attempt and the methods used to achieve their goals. Finally, we point out several understudied areas in this field and make suggestions for future directions.
