Kidney disease models: tools to identify mechanisms and potential therapeutic targets

  • 摘要: 近年来,急性肾损伤(AKI)和慢性肾脏病(CKD)的增患病率快速增长,已经成为影响全球数百万人的公共卫生问题。由于导致肾功能衰竭的病因具有多样性,因此不同的动物模型被开发出来以满足对不同病理机制的研究,促进我们对肾脏病的理解。在这些实验模型中,啮齿类动物模型被广泛用于系统性地研究肾脏疾病的诱因和发生发展,以及探索潜在治疗靶点。本综述旨在讨论和总结使用手术或药物、毒素诱导的手段进行AKI和CKD动物建模的方法,以及应用各种转基因小鼠进行CKD建模的方法。此外,本综述还探讨了近期肾脏疾病相关临床试验的结果和动物模型相关的新兴概念。

     

    Abstract: Acute kidney injury (AKI) and chronic kidney disease (CKD) are worldwide public health problems affecting millions of people and have rapidly increased in prevalence in recent years. Due to the multiple causes of renal failure, many animal models have been developed to advance our understanding of human nephropathy. Among these experimental models, rodents have been extensively used to enable mechanistic understanding of kidney disease induction and progression, as well as to identify potential targets for therapy. In this review, we discuss AKI models induced by surgical operation and drugs or toxins, as well as a variety of CKD models (mainly genetically modified mouse models). Results from recent and ongoing clinical trials and conceptual advances derived from animal models are also explored.
